چکیده

The interferons λ (IFNsλ), also known as IL-28 and IL-29, are co-expressed with IFNβ following Toll Like Receptor (TLR) stimulation in human monocyte-derived dendritic cells (DCs). IFNλ shares with type I IFNs an intracellular signaling pathway which drives the expression of a common set of gene. However, IFNλ signaling is initiated through a membrane receptor system distinct from that of type I IFNs. Because IFNs produced by DCs in response to TLR stimulation are critical in both differentiation and maturation of DCs, we sought to investigate whether IFNλ exhibits specific effects on DCs differentiation. We show in this work that DCs acquire IFNλ responsiveness through the expression of the specific IFNλ receptor chain during their differentiation from monocytes. IFNλ-treated DCs express high levels of MHC I and II but low levels of costimulatory molecules. However, they express CCR7 and acquire the ability to migrate to lymph nodes when intravenously injected into SCID/Bg mice. In MLR cultures, IFNλ-treated DCs specifically induced an IL-2dependent proliferation of a CD4, CD25, Foxp3 T cell subset with contact-dependent suppressive activity on T cells proliferation initiated by fully mature DCs. IFNλ are thus able to generate tolerogenic DCs, an activity that could thwart IFNβ functions. For personal use only. on November 12, 2017. by guest www.bloodjournal.org From

Effect of Mesenchymal Stem Cells on ILT3 Expression in the Splenocytes of Skin Graft Recipient Mice

Background: Mesenchymal stem cells (MSCs) are considered as effective therapeutic cells in transplantation due to their immunomodulatory activities. However, precise mechanism of MSCs immunomodulatory activity is not completely understood. Objectives: To study the role of Immunoglobulin-like transcripts-3 (ILT3) immunomodulatory receptor in immune tolerance induced by MSCs in skin transpla...

CD40 Knocked Down Tolerogenic Dendritic Cells Decrease Diabetic Injury

Background: Type-1 diabetes (T1D) is an autoimmune disease in which T lymphocytes destroy insulin-producing β-cells. Control of self-reactive T lymphocytes and recovery of diabetic injury is the end point of T1D. Objective: To investigate generation of tolerogenic dendritic cells (tolDCs) as an innovative method of diabetes therapy. Methods: Lentivirus vector production was achieved by GIPZ mou...

بررسی تأثیر سرم موش حامله بر روی سلولهای دندریتیک در القاء تحریک لنفوسیت‌های T و تولید سیتوکین‌های IL-10 و IFN-γ Dendritic Cells and Antigen Specific T Cell Responses: Effect of Pregnant Mouse Serum

Background & Aim: Tolerance to the semi-allogenic fetal graft by the maternal immune system is a medical enigma that has stimulated investigations for a half of century. Several hypotheses have been proposed to explain the tolerance of mother to the fetus. The successful pregnancy is proposed and proved by many scientists to be a Th2 dominant phenomenon. This hypothesis is proved in most as...

TGF-Β1 Transduced Mesenchymal Stem Cells Have Profound Modulatory Effects on DCs and T Cells

Background: Mesenchymal stem cells (MSCs) and transforming growth factor-β1 (TGF-β1) molecules are well known for their immunomodulatory properties and their function in tissue regeneration and remodeling. Objectives: To evaluate the interaction of TGF-β1 engineered MSCs with T cells and dendritic cells (DCs) and their modulatory effect on the immune response. Methods: MSCs and DCs were generat...

Bone marrow dendritic cells deficient for CD40 and IL-23p19 are tolerogenic in vitro

Objective(s): In addition to pro-inflammatory role, dendritic cells (DCs) can also be anti-inflammatory when they acquire tolerogenic phenotype. In this study we tested the role of CD40 and IL-23p19 in antigen presenting function of bone marrow-derived DCs (BMDCs) by comparing BMDCs derived from CD40 knockout (CD40KO-DCs) and IL-23p19 (IL-23p19KO-DCs) knockout mice wit...
